Allopatric chromosomal variation in Nematocharax venustus Weitzman, Menezes & Britski, 1986 (Actinopterygii: Characiformes) based on mapping of repetitive sequences Neotropical Ichthyology
Barreto,Silvia B.; Cioffi,Marcelo B.; Medrado,Aline S.; Silva,André T.; Affonso,Paulo R. A. M.; Diniz,Débora.
ABSTRACT Characiformes is the most cytogenetically studied group of freshwater Actinopterygii, but karyotypical data of several taxa remain unknown. This is the case of Nematocharax , regarded as a monotypic genus and characterized by marked sexual dimorphism. Therefore, we provide the first cytogenetic report of allopatric populations of Nematocharax venustus based on distinct methods of chromosomal banding and fluorescence in situ hybridization (FISH) with repetitive DNA probes (18S and 5S rDNA). The karyotype macrostructure was conserved in all specimens and populations, independently on sex, since they shared a diploid number (2n) of 50 chromosomes divided into 8m+26sm+14st+2a. The heterochromatin was mainly distributed at pericentromeric regions and...

B chromosome and NORs polymorphism in Callichthys callichthys (Linnaeus, 1758) (Siluriformes: Callichthyidae) from upper Paraná River, Brazil Neotropical Ichthyology
Konerat,Jocicléia Thums; Bueno,Vanessa; Baumgartner,Lucas; Martins-Santos,Isabel Cristina; Margarido,Vladimir Pavan.
B chromosomes are extra chromosomes from the normal chromosomal set, found in different organisms, highlighting their presence on the group of fishes. Callichthys callichthys from the upper Paraná River has a diploid number of 56 chromosomes (26 m-sm + 30 st-a) for both sexes, with the presence of a sporadically acrocentric B chromosome. Moreover, one individual presented a diploid number of 57 chromosomes, with the presence of a morphologically ill-defined acrocentric B chromosome in all analyzed cells. The physical mapping of 5S and 18S rDNA shows multiple 5S rDNA sites and only one pair of chromosomes with 18S sites in C. callichthys, except for two individuals. These two individuals presented a third chromosome bearing NORs (Ag-staining and 18S rDNA)...

Bonamia exitiosus n. sp (Haplosporidia) infecting flat oysters Ostrea chilensis in New Zealand ArchiMer
Hine, P; Cochennec, Nathalie; Berthe, Franck.
Bonamia sp. is a pathogenic parasite that occurs in the haemocytes of dredge oysters Ostrea chilensis Philippi in New Zealand, Ultrastructurally it resembles other haplosporidians in the possession of haplosporosomes, haplosporogenesis, persistence of mitotic microtubules during interphase and of the nuclear envelope during mitosis, and occurrence of a diplokaryotic or multi-nucleate plasmodial stage. Another stage containing a large vacuole derived from enlargement of 1 or more mitochondria has not previously been described from other haplosporidians. It most closely resembles B, ostreae Pichot et al., 1979, which parasitises and is pathogenic in haemocytes of European flat oysters, O. edulis. However, B. ostreae is smaller and denser, and has fewer...

Comparative cytogenetic analyses in Ancistrus species (Siluriformes: Loricariidae) Neotropical Ichthyology
Glugoski,Larissa; Deon,Geize; Schott,Stephane; Vicari,Marcelo R.; Nogaroto,Viviane; Moreira-Filho,Orlando.
ABSTRACT Ancistrus is a specious genus of armored catfishes that has been extensively used for cytogenetic studies in the last 17 years. A comparison of the extensive karyotypic plasticity within this genus is presented with new cytogenetic analysis for Ancistrus cf. multispinis and Ancistrus aguaboensis. This study aims to improve our understanding of chromosomal evolution associated with changes in the diploid number (2n) and the dispersion of ribosomal DNAs (rDNAs) within Ancistrus. Ancistrus cf. multispinis and A. aguaboensis exhibit 2n of 52 and 50 chromosomes, respectively. Given that A. cf. multispinis shares a 2n = 52 also found in Pterygoplichthyini, the sister group for Ancistrini, a Robertsonian (Rb) fusion event is proposed for the 2n reduction...

Contributions to the systematic of Pimelodidae (Osteichthyes, Siluriformes): basic and molecular cytogenetics on seven species of Pimelodus from three Brazilian hydrographic systems Neotropical Ichthyology
Girardi,Simone C.; Pavanelli,Carla S.; Margarido,Vladimir P..
ABSTRACT Pimelodidae harbors several species and is widely distributed throughout the Neotropical region. Pimelodus is the genus with the largest number of species, however it is a polyphyletic group. Cytogenetic analyzes of the valid species still covers less than half of them. Herein, seven Pimelodus species from three Brazilian hydrographic systems were analyzed through basic (Giemsa, AgNORs and C banding) and molecular (5S and 18S rDNA-FISH) cytogenetic methods. All species had 2n=56 chromosomes with different karyotype formulas observed among the species. AgNORs were corresponding to 18S rDNA and localized on long arm of one chromosome pair in all species. Heterochromatin distribution follows the pattern commonly verified in the family and allows to...

Cytogenetic analysis of Astylus antis (Perty, 1830) (Coleoptera, Melyridae): Karyotype, heterochromatin and location of ribosomal genes Genet. Mol. Biol.
Mendes-Neto,Ernani de Oliveira; Vicari,Marcelo Ricardo; Campaner,Carlos; Nogaroto,Viviane; Artoni,Roberto Ferreira; Almeida,Mara Cristina.
Cytogenetic analysis of Astylus antis using mitotic and meiotic cells was performed to characterize the haploid and diploid numbers, sex determination system, chromosome morphology, constitutive heterochromatin distribution pattern and chromosomes carrying nucleolus organizer regions (NORs). Analysis of spermatogonial metaphase cells revealed the diploid number 2n = 18, with mostly metacentric chromosomes. Metaphase I cells exhibited 2n = 8II+Xyp and a parachute configuration of the sex chromosomes. Spermatogonial metaphase cells submitted to C-banding showed the presence of small dots of constitutive heterochromatin in the centromeric regions of nearly all the autosomes and on the short arm of the X chromosome (Xp), as well as an additional band on one of...

Cytogenetic and molecular markers reveal the complexity of the genus Piabina Reinhardt, 1867 (Characiformes: Characidae) Neotropical Ichthyology
Pazian,Marlon Felix; Pereira,Luiz Henrique Garcia; Shimabukuru-Dias,Cristiane Kioko; Oliveira,Claudio; Foresti,Fausto.
Cytogenetic and molecular analyses were carried out in fish representative of the genus Piabina. This study specifically involved the species P. argentea and P. anhembi collected from areas of the Paranapanema and Tietê River basins, Brazil. Our findings suggest that fish classified as Piabina argentea in the Paranapanema and Tietê Rivers may represent more than one species. The samples analyzed differed by cytogenetic particularities and molecular analyses using partial sequences of the genes COI and CytB as genetic markers revealed three distinct groups of P. argentea with genetic distances sufficient to support the conclusion that the three samples analyzed are three distinct taxonomic units.

Cytogenetic data on Astyanax jacuhiensis (Characidae) in the lago Guaíba and tributaries, Brazil Neotropical Ichthyology
Pacheco,Rosiley B.; Giuliano-Caetano,Lucia; Julio Junior,Horácio F.; Dias,Ana L..
Cytogenetic analyses were performed in Astyanax jacuhiensis from lago Guaíba, Brazil. The diploid number was 50, with a karyotype composed of 8m+30sm+4st+8a chromosomes, FN = 92. The AgNORs were observed in 2 to 5 chromosomes, with intra- and interindividual variation. The sm pair 8 observed always carried NORs on the short arms, presenting size heteromorphism between homologous. Fluorescence in situ hybridization (FISH) with an 18S rDNA probe only confirmed the location of ribosomal cistrons in the sm pair 8, and heteromorphism of these regions between the homologous chromosomes. C-banding revealed the occurrence of weak C-positive heterochromatin in the pericentromeric regions of several chromosomes, in addition to more evident bands interstitially...

Cytotaxonomy of Parodon nasus and Parodon tortuosus (Pisces, Characiformes): A case of synonymy confirmed by cytogenetic analyses Genet. Mol. Biol.
Bellafronte,Elisangela; Margarido,Vladimir Pavan; Moreira-Filho,Orlando.
Morphological and cytogenetical studies were carried out on the freshwater fish Parodon nasus and Parodon tortuosus in order to evaluate a putative synonymy. The diploid chromosome number observed in both species was 2n = 54 (48M/SM and 6ST) with no differences between the sexes. Despite slight differences in the pattern of heterochromatin distribution and the number of cusps in symphysean teeth, the location of nucleolar organizer regions (NORs) and 5S rRNA genes (both species-specific features) were similar in both species. The remarkable similarity observed between these allopatric species supports recent taxonomical reviews indicating that P. tortuosus is a synonym for P. nasus.

Extensive polymorphism and chromosomal characteristics of ribosomal DNA in the characid fish Triportheus venezuelensis (Characiformes, Characidae) Genet. Mol. Biol.
Nirchio,Mauro; Oliveira,Claudio; Ferreira,Irani Alves; Granado,Angel; Ron,Ernesto.
The karyotype and chromosomal characteristics of the characid fish Triportheus venezuelensis were investigated using differential staining techniques (C-banding, Ag-NOR staining) and fluorescent in situ hybridization (FISH) with an 18S rDNA probe. The diploid chromosome number (2n = 52), karyotype composition and sex chromosome determination system of the ZZ/ZW type were the same as previously described in other species of the genus Triportheus. However, extensive variation regarding nucleolus organizer regions (NOR) different from other species was observed. 18S rDNA sequences were distributed on nine chromosome pairs, but the number of chromosomes with Ag-NORs was usually lower, reaching a maximum of four chromosomes. When sequential staining experiments...

Karyotypic conservatism in samples of Characidium cf. zebra (Teleostei, Characiformes, Crenuchidae): physical mapping of ribosomal genes and natural triploidy Genet. Mol. Biol.
Pansonato-Alves,José Carlos; Oliveira,Claudio; Foresti,Fausto.
Basic and molecular cytogenetic analyses were performed in specimens of Characidium cf. zebra from five collection sites located throughout the Tietê, Paranapanema and Paraguay river basins. The diploid number in specimens from all samples was 2n = 50 with a karyotype composed of 32 metacentric and 18 submetacentric chromosomes in both males and females. Constitutive heterochromatin was present at the centromeric regions of all chromosomes and pair 23, had additional interstitial heterochromatic blocks on its long arms. The nucleolar organizer regions (NORs) were located on the long arms of pair 23, while the 5S rDNA sites were detected in different chromosomes among the studied samples. One specimen from the Alambari river was a natural triploid and had...

Karyotypic evolution of ribosomal sites in buffalo subspecies and their crossbreed Genet. Mol. Biol.
Degrandi,Tiago Marafiga; Pita,Sebastian; Panzera,Yanina; Oliveira,Edivaldo Herculano C. de; Marques,José Ribamar Felipe; Figueiró,Marivaldo Rodrigues; Marques,Larissa Coêlho; Vinadé,Lucia; Gunski,Ricardo José; Garnero,Analía Del Valle.
Domestic buffaloes are divided into two group based on cytogenetic characteristics and habitats: the "river buffaloes" with 2n = 50 and the "swamp buffaloes", 2n = 48. Nevertheless, their hybrids are viable, fertile and identified by a 2n = 49. In order to have a better characterization of these different cytotypes of buffaloes, and considering that NOR-bearing chromosomes are involved in the rearrangements responsible for the karyotypic differences, we applied silver staining (Ag-NOR) and performed fluorescent in situ hybridization (FISH) experiments using 18S rDNA as probe. Metaphases were obtained through blood lymphocyte culture of 21 individuals, including river, swamp and hybrid cytotypes. Ag-NOR staining revealed active NORs on six chromosome pairs...

Localization of ribosomal genes in three Pimelodus species (Siluriformes, Pimelodidae) of the São Francisco River: 5S genes as species markers and conservation of the 18S rDNA sites Genet. Mol. Biol.
Garcia,Caroline; Moreira Filho,Orlando.
Pimelodidae is one of the most representative of Neotropical catfish families. However, these fish are still poorly studied in terms of cytogenetics, especially regarding the application of more accurate techniques such as the chromosomal localization of ribosomal genes. In the present work, fluorescent in situ hybridization with 5S and 18S rDNA probes was employed for rDNA site mapping in Pimelodus sp., P. fur and P. maculatus from the São Francisco River in the Três Marias municipality - MG. The results from the application of the 18S probe confirmed the previous data obtained by silver nitrate staining, identifying a simple nucleolar organizing region system for these species. However, the labeling results from the 5S rDNA probe demonstrated a...

Mapping of the 18S and 5S ribosomal RNA genes in Astyanax altiparanae Garutti & Britski, 2000 (Teleostei, Characidae) from the upper Paraná river basin, Brazil Genet. Mol. Biol.
Fernandes,Carlos Alexandre; Martins-Santos,Isabel Cristina.
Fluorescence in situ hybridization (FISH) was undertaken in order to determinate the chromosomal distribution pattern of 18S and 5S ribosomal DNAs (rDNA) in four populations of the characid fish Astyanax altiparanae from the upper Paraná river basin, Brazil. The 18S rDNA probe FISH revealed numerical and positional variations among specimens from the Keçaba stream compared to specimens of the other populations studied. In contrast to the variable 18S rDNA distribution pattern, highly stable chromosomal positioning of the 5S rDNA sites was observed in the four A. altiparanae populations. Divergence in the distribution pattern of 18S and 5S rDNA sites is also discussed.

NA phylogeny of Ryukyus Leucothoidae (Crustacea: Amphipoda) Naturalis
White, K.N.; Reimer, J.D..
Commensal leucothoid amphipods collected from sponges, ascidians, and coral rubble from the Ryukyu Archipelago, Japan, were investigated using nuclear 18S ribosomal DNA sequences. Analysis of sequences from 21 species in three genera supported the current morphological species designations and the separation of the family into two clades. Additionally, a possible new generic-level clade was identified and the separation of the genera Anamixis and Paranamixis was not supported. Our results demonstrate that host specificity appears to evolve rapidly in sibling species in this family.

Phylogeny and evolution of Burmanniaceae (Dioscoreales) based on nuclear and mitochondrial data Naturalis
Merckx, V.; Schols, P.; Maas - Van de Kamer, H.; Maas, P.; Huymans, S.; Smets, E..
The mycoheterotrophic Burmanniaceae are one of the three families currently recognized in the order Dioscoreales. Phylogenetic inference using nucleotide sequences of the nuclear 18S rDNA region and the mitochondrial nad1 b-c intron revealed two well-supported, major lineages within the family, corresponding to the two tribes recognized in the family: Burmannieae and Thismieae. All data supported a strong relationship between Thismieae and Tacca (Dioscoreaceae) making both Burmanniaceae and Dioscoreaceae polyphyletic. The three largest Burmanniaceae genera, Burmannia, Gymnosiphon, and Thismia, are paraphyletic. The splitting of Burmanniaceae into Burmannieae and Thismieae indicates two independent origins of mycoheterotrophy and correlated loss of...

Sex chromosome system ZZ/ZW in Apareiodon hasemani Eigenmann, 1916 (Characiformes, Parodontidae) and a derived chromosomal region Genet. Mol. Biol.
Bellafronte,Elisangela; Schemberger,Michelle Orane; Artoni,Roberto Ferreira; Moreira Filho,Orlando; Vicari,Marcelo Ricardo.
Parodontidae fish show few morphological characteristics for the identification of their representatives and chromosomal analyses have provided reliable features for determining the interrelationships in this family. In this study, the chromosomes of Apareiodon hasemani from the São Francisco River basin, Brazil, were analyzed and showed a karyotype with 2n = 54 meta/submetacentric chromosomes, and a ZZ/ZW sex chromosome system. The study revealed active NORs located on pair 11 and additional 18S rDNA sites on pairs 7 and 22. The 5S rDNA locus was found in pair 14. It showed a pericentric inversion regarding the ancestral condition. The satellite DNA pPh2004 was absent in the chromosomes of A. hasemani, a shared condition with most members of Apareiodon....

Towards a phylogeny of the Metazoa: evaluating alternative phylogenetic positions of Platyhelminthes, Nemertea, and Gnathostomulida, with a critical reappraisal of cladistic characters Naturalis
Jenner, Ronald A..
This paper critically assesses all morphological cladistic analyses of the Metazoa that were published during the last one and a half decades. Molecular and total evidence analyses are also critically reviewed. This study focuses on evaluating alternative phylogenetic positions of the ‘acoelomate’ worms: Platyhelminthes, Nemertea, and Gnathostomulida. This paper consists of two parts. In Part I, all recently proposed sister group hypotheses and the supporting synapomorphies for these phyla are evaluated. Discrepancies in the treatment of corresponding characters in different cladistic analyses are identified, and where possible, resolved. In Part II, the overall phylogenetic significance across the Metazoa of all characters relevant for placing the...
